ABSTRACT

We analyzed the pathogenic characteristics of Salmonella strains isolated from diarrhea patients in Wuxi City,Jiangsu Province,China and compared the differences among pulse field gel electrophoresis (PFGE) patterns of main serotype strains,so as to provid scientific basis for disease control.After biochemical identification of the Salmonella strains isolated from infectious diarrhea patients in Wuxi in 2015,drug susceptibility test,serotyping and PFGE were applied to analyze these strains.Results showed that a total of 32 Salmonella strains were detected from 756 diarrhea specimens with a positive rate of 4.23 %.The infection occurred more frequently between May and October and adults aged more than 60 years old affected mostly.There was no significant difference between genders in infected population.The drug susceptibility test indicated that the antibiotic resistance rate of these Salmonella strains to ampicillin (56.25 %) was the highest,and to ciprofloxacin(6.25 %)and Ceftazidime (6.25%) were the lowest.The 32 Salmonella strains belonged to 11 serotypes,and S.enteritidis(31.25%)and S.typhimurium(21.88%) were the predominant serotypes.PFGE showed that the pattern similarity of all S.enteritidis was more than 85 %;PFGE patterns of S.typhimurium were different.In conclusion,the infection of Salmonella from diarrhea patients in Wuxi City had obvious season and age specific distribution,and the most prevalent serotype of Salmonella was the S.enteritidis.It is necessary to strengthen the surveillance of Salmonella concurrently in food and environment.

ABSTRACT

Objective To study the effects of microemulsion/ethosomes on transdermal absorption properties and efficacy of Huoxue Zhitong Cataplasm. Methods The improved Franz diffusion cells were used for the in-vitro permeation experiment with rat skins as the barriers, which was used to evaluate the transdermal absorption properties. In the erxeriment, the contents of paeonol, eugenol and methyl salicylate were used as markers, and detected by ultra performance liquid chromatography to evaluate the transdermal absorption effects. The anti-inflammatory and analgesia activity were evaluated through the writhing plate experiments. Results The cumulative release rate of paeonol in Huoxue Zhitong Cataplasm, Microemulsion Huoxue Zhitong Cataplasm and Ethosomes Huoxue Zhitong Cataplasm were, in order, 65.30%, 61.30%and 60.20%in 24 h;eugenol were, in order, 51.08%, 54.71% and 55.66% in 24 h; methyl salicylate were, in order, 49.20%, 65.17% and 72.15% in 24 h. Furthermore, Microemulsion Huoxue Zhitong Cataplasm high-dose group and Ethosomes Huoxue Zhitong Cataplasm medium-dose group had good effects on reducing the inflammatory exudate of peritoneal capillary and capillary permeability (P<0.05) in animal models. Conclusion Huoxue Zhitong Cataplasm based on microemulsion/ethosomesnano-technology has good transdermal absorption properties and efficacy.

ABSTRACT

ObjectiveTo develop a quantitative method for simultaneously determining multi-components in Rhei Radix et Rhizoma using one chemical reference substance.MethodsThe contents of multi-components were calculated by the UV relative correction factors (RCFs) of chrysophanol,physcion,and rhein to emodin.ResultsThe values of RCFs at 274 nm for rhein,chrysophanol,and physcion to emodin were 0.712,0.674,and 1.051.The calibration curves were linear over the ranges of 0.02-4.08,0.02-4.12,0.07- 12.92,and 0.02-3.68 μg/mL for rhein,emodin,chrysophanol,and physcion,respectively.The contents of emodin in 18 samples were determined by the extemal standard method,and the contents of the other three anthraquinone aglycones were calculated according to their RCFs.ConclusionNo significant difference is found in comparison with the classical method,indicating that the RCFs have high reliability within their linear ranges and could be used in quality control of Rhei Radix et Rhizoma.The quantitative analysis of multi-component with a single marker is especially suitable for herbal medicines containing unstable or hard to be purified components as quality control markers.

ABSTRACT

Objective To investigate the effects of a new c-Met inhibitor SU11274 on apoptosis and motility of c-Met-positive basal-like breast cancer cells MDA-MB-231. Methods The concentrations of SUl1274 were set to 0,0.1,1,10 and 20 μmol/L.Morphological change of apoptotic cells was analyzed by Hoechst33342,MitroTrackerRed and Yo-pro-1 staining.The apoptotic rate of MDA-MB-231 cells were determined by Annexin V/PI double-staining. The expression of apoptosis related proteins (Bcl-XL,Caspase-3 and PARP) and phosphorylation levels of c-Met and Akt were analyzed by Western blot.The capability of motility were measured by wound-healing assay and chemotaxis assay. Results After treatment by SU11274( 10 μmol/L) for 48 h,shrinking apoptotic cells of MDA-MB-231 was observed by flurescent microscope and nuclear fragmentation was seen.Annexin V/PI double-staining showed SU11274induced apoptosis of MDA-MB-231 cells (P < 0.05 ),and the apoptotic rates were (7.3 ± 0.9) %,( 14.1 ±0.6) %,(35.5 ± 4.4) % and (48.2 ± 5.3 ) %,respectively.SU11274 downregulated the expression of Bcl-XL and promoted the dissection of Caspase-3 and PARP in a dose dependent relationship.SU11274 prolongs the wound-healing time,decreases the migration cell count (P < 0.05 ) and effectively inhibits the phosphorylation of c-Met and its downstream key proteins Akt in a dose-dependent manner.Conclusions C-Met inhibitor SU11274 induces apoptosis and inhibits the motility of c-Met-positive basallike breast cancer cell line MDA-MB-231,probably through inhibiting phosphorylation of c-Met/PI3K/Akt.

ABSTRACT

<p><b>OBJECTIVE</b>To establish a quality evaluation method, quantitative analysis of multi-component with a single-marker (QAMS) to determine the contents of four rhubarb anthraquinones in Sanhuang tablets.</p><p><b>METHOD</b>Emodin was used as the internal reference substance, the relative correlation factors (RCF) of rhein, chrysophanol and physcion to emodin were calculated and evaluated. The contents of these four anthraquinones were determined by the external standard method and QAMS, respectively. Rationality, feasibility and repeatability of the QAMS method was verified by comparing the results obtained from the two different methods.</p><p><b>RESULT</b>RCFs of rhein, chrysophanol and physcion to emodin against Sanhuang tablets were 1.13, 1.46 and 1.01, respectively, indicating good reproducibilities. The two methods did not show significant difference in results.</p><p><b>CONCLUSION</b>The QAMS method can be used a new quality assessment model for quantity of anthraquinones in Sanhuang tablets.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To examine the feasibilities of the method, quantitative analysis of multi-components by single marker (QAMS) for determination of four alkaloids in Xianglian preparations.</p><p><b>METHOD</b>A HPLC method was developed to determine the contents of four alkaloids, epiberberine, coptisine, palmatine and berberine in Xianglian preparations. Berberine was selected as the internal reference substance, and 3 relative correction factors to berberine were calculated. The contents of the four alkaloids in 16 batches of samples were determined by both external standard method and QAMS. The new method was evaluated by comparing the quantitative results with of the external standard method.</p><p><b>RESULT</b>No significant differences were found between the quantitative results of four alkaloids in 16 batches of samples determined by the two methods.</p><p><b>CONCLUSION</b>QAMS is feasible for determination of four alkaloids simultaneously, and the developed method can be used for quality control of Xiang Lian preparations.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To establish a new quantitative method for simultaneous determination of multi-coumarins in Fraxini Cortex by using one chemical reference substance, and validate its feasibilities.</p><p><b>METHOD</b>The new quality evaluation method, quantitative analysis of multi-components by singer-marker (QAMS), was established and validated with Fraxini Cortex. Four main coumarins were selected as analytes to evaluate the quality and their relative correlation factors (RCF) were determined by HPLC-DAD. Within the linear range, the values of RCF at 340 nm of aesculin to asculetin, fraxin and fraxetin were 1.771, 0.799, 1.409, respectively. And the contents of aesculin in samples of Fraxini Cortex were authentically determined by the external standard method, and the contents of the three other coumarins were calculated by their RCF. The contents of these four coumarins in all samples were also determined by the external standard method.</p><p><b>RESULT</b>Within a certain range, the RCF had a good reproducibility (RSD 2.5%-3.9%). Significant differences were not observed between the quantitative results of two methods.</p><p><b>CONCLUSION</b>It is feasible and suitable to evaluate the quality of Fraxini Cortex and its Yinpian by QAMS.</p>

ABSTRACT

Epithelial-mesenchymal transition cause primary carcinoma cells to acquire mesenchymal features and re-epithelialize to form a secondary mass at a metastatic site. Such plasticity has implications in the progression of breast carcinoma to metastasis, and will likely influence cancer's response to therapy. The transcriptional and epigenetic regulation that underlie the development of breast cancer and result in characteristic changes in cell behavior can be monitored using an array of marker proteins, providing the potential for emergent prognostic and therapeutic targeting.

ABSTRACT

<p><b>OBJECTIVE</b>To establish a new quantitative method for simultaneous determination of multi-components in Salvia miltiorrhiza by using one chemical reference substance and validate its feasibilities.</p><p><b>METHODS</b>The new quality evaluation method, quantitative analysis of multi-components by singer-marker (QAMS), was established and validated with S. miltiorrhiza . Five main ingredients were selected as analytes to evaluate the quatity and their relative correlation factors (RCF) were determined by HPLC-DAD. Within the linear range, the values of RCF at 280 nm of sodium danshensu to protocatechuic aldehyde, rosmarinic acid, lithospermic acid and salvianolic acid B were 7.559, 2.901, 1.825 and 1.746, respectively. The contents of danshensu in the samples of S. miltiorrhiza were determined by using the external standard method, and the contents of the four other ingredients were calculated by their RCFS. The contents of these four ingredients in all samples were also determined by the external standard method.</p><p><b>RESULT</b>Within a certain range, the RCFs showed good reproducibility (RSD 1.0%-2.5%). No significant differences were observed between the quantitative results of the two methods.</p><p><b>CONCLUSION</b>It is feasible and suitable to evaluate the quality of S. miltiorrhiza and its processed products by QAMS.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To develop a Quantitative Assay of Multi-components by Single - marker (QAMS) for simultaneous determination of three components in Fructus Evodiae, and examine the feasibility of using the relative correction factors between the different types of compounds.</p><p><b>METHOD</b>Rutaecarpine was selected as the internal reference substance; the relative correction factors of evodin and evodiamine were calculated. The contents of three components in 11 batches of samples were determined by both external standard method and QAMS. The validity of the QAMS method was evaluated by comparison of their quantitative results.</p><p><b>RESULT</b>No obvious differences (RSD < 5%) were found in the quantitative results of evodin and evodiamine in 11 batches of Fructus Evodiae determined by the two methods.</p><p><b>CONCLUSION</b>It is feasible and suitable to determine evodin and evodiamine in Fructus Evodiae by QAMS, and this method can be used for a certain different types of compounds.</p>

ABSTRACT

Objective To establish a method for determination of the water-soluble components of Salvianolic acid B,Danshensu and Protocatechuic aldehyde in Salvia miltiorrhiza Bge. Method Chromatography was performed on Kromosil C18 (4.6 mm?250 mm,5 ?m). Mobile phase Ⅰ was methanol-acetonitrile-formicacid-water (30∶10∶1∶59),mobile phase Ⅱ was methanol-acetic acid-water (20∶1∶80),the flow rate was 1.0 mL/min,the detection wavelength was set at 286 and 280 nm,column temperature was 35 ℃. Result Determined 10 different habitats Salvia miltiorrhiza Bge.,the average content of Salvianolic acid B was 0.09%~6.81%,the average recovery was 99.64%,RSD=1.09%. The average content of Danshensu was 0.16%~0.68%,the average recovery was 98.94%,RSD=1.49%. The average content of Protocatechuic aldehyde was 0.01%~0.09%,the average recovery was 98.96%,RSD= 1.66%. Conclusion The method is accurate and reliable with good reproducibility,and can be used as determination standard for Salvia miltiorrhiza Bge.

ABSTRACT

The recent advances in research of oxygen free radicals and its relation to Parkinsons disease (PD) was reviewed. Though there is no exact knowledge of the causes of PD, the role of oxidative stress in the etiology of PD has been focused on. Investigations on patients have shown that PD is under a status of oxidative stress. The biochemical features of dopaminergic neurons in the pathologic site of PD implicate the close relation of oxygen free radicals' generation with the pathogenesis of PD. Besides the oxidative stress hypothesis, other proposed hypotheses are also direct or indirect related to oxidative stress.
